In SCIEX OS software, internal standard (IS) recovery can be determined using a custom calculation in the quantitation method. When creating a quantitation method, select the Calculated Columns option and then select Add Formula to bring up the Custom Calculation interface.

To calculate IS recovery, input the following formula: [IS Area]/MEAN([IS Area]). Selecting MEAN for the average allows you to choose which types of samples to use in the calculation.

In this case, only standards should be checked.

Select Accept changes and return to calculated columns to save the formula. When data is processed using this method, a custom column will be generated in the results table that displays the IS recovery value.

MultiQuant™ software does not have a feature to build calculated columns; instead, a query must be applied to the results table to generate a custom column that appears in the results table with the values for IS recovery. The query can be provided by reaching out to SCIEX tech support and then placed into the following location: C:\ProgramData\AB SCIEX\MultiQuant\Queries. To run a query in MultiQuant software, select the Query option in the Process menu.

The results table will be updated with a % Recovery column.
